was after so tips on shooting real close-ups/macro shots of eyes. I wouldn't know where to start not so flash at people.

Why not experiment?
If you prefer not to use flash, the other suggestion I'd have is to crank your 350D up to ISO 1600 or so and try shooting at f/4 or f/5.6 in fairly bright light with your 100mm macro. You could use a desk lamp or bright, concentrated lamp to get more light onto the eye. Shooting in RAW will give you the ability to correct any color cast in the light.
All this assumes a willing subject and controlled conditions, which I assume is what you intend!

Thanks for that, the subject is my wife and she is patient. I want to use macro lense and thought about using my twinlight flash dialed rightdown this flash should avoid red-eye
